11 Analytical Procedures

12 Quality Assurance and Quality Control (QA/QC) Checks

12.1 Confmnation and verification of presumed oocysts and/or cysts are perfonned by making comparison to the positive and negative control organisms examined with each sample. Cysts and oocysts are only "confinned" if internal stnictures are identified. 12.2 As a sterility indicator, the temperature is checked during autoclaving runs to make sure appropriate sterilization temperatures are being attained. This is maintained in the logbook that is kept near the autoclave. The accurateness of the autoclave is checked by autoclave tape, kill-it ampules, broth tests and temperature checks. These results are recorded in a logbook located next to the autoclave. 12.3 Over the course of a week, all of the samples processed using the ICR method are considered a BATCH. A negative and a positive filter are processed as QC samples for each batch for each week. These filters are prepared according to the instructions in the ICR for QC samples. Matrix spikes are perfoiTned weekly and matrix spike duplicates are perfonned monthly. 12.4 Over the course of a week, all of the samples processed using the 1623 method ai’e considered a BATCH. A negative and a positive filter are processed as QC samples for each batch for each week. These filters are prepared according to the instructions in the 1623 manual for QC samples. Matrix spikes are perfonned weekly. 12.5 The PE samples are cunently distributed by the US EPA, since NYC DEP is part of the National Performance Evaluation Program.

12.6 Deviation in microscopic counts between analysts is derived from duplicate counts perfonned by each Principle Analyst quarterly.
