9.1.2 Chemicals
9.1.2.1 One bottle of each of the chemicals used in making the solutions are kept
in a storage cabinet.
9.1.2.2 All of the unopened bottles are kept in chemical storage in the basement
of Ben Nesin Lab. Such chemicals are sucrose, sodium chloride, laurel
sulfate, sodium phosphate, potassium chloride, tween 80, glycerol, and
potassium phosphate.
9.1.2.3 Other chemicals are stored under refrigeration such as: bovine albumin
and percoll. The supply of formaldehyde is kept in the lab facility's
chemical storage area.
9.1.2.4 Chemicals are dated with the following; date received, date opened and
expiration date.
9.1.2.5 The specifications for the chemicals that are used for making reagents,
are the same as what is stated in the Information Collection Rule.
9.1.3 Filters
9.1.3.1 Two types of filters are used in the Hoefer Manifold procedure. The
bottom or support filter is a cellulosic, 5.0 micron, 25mm filter. The top
filter is a cellulose acetate 0.22 micron, 25 mm filter. These filters are
stored in a bench drawer, in plastic storage containers, when not in use.
9.1.3.2 Gelman Envirochek Capsules are used in the ERA method 1623
procedure. These are approximately 6 cm in diameter and 21 cm long
containing a polyethersulfone filter media and inlet and outlet fittings.
These filters are stored in a cabinet outside the Pathogen Lab door.
10 Laboratory Practices
10.1 Reagent-grade water is prepared using a Mill-Q Water System, MRO system,
manufactured by the Millipore Co.
10.2 After glassware is used it is put on the counter next to the sink to be washed. The
diity glassware is either washed with a diluted Micro cleaning solution or a
diluted Coverage Plus Brand disinfectant and cleaner. After the glassware is
washed, it is laid out to air diy and then put away in the appropriate storage
places. Prior to beginning analysis, the designated glasswai-e is laid out on the
workbench and labeled with the appropriate solution names, sample numbers and
appropriate dates. This is done to avoid any confusion during the sample
processing procedure.
10.3 All glassware use in the 1623 procedure is autoclaved, scmbbed with a wann
detergent solution and then soaked in a 5% hypochlorite solution for a minimum
of 30 minutes. Then the glassware is rinsed with reagent water and placed out to
diy.
10.4 Disposable supplies ai'e used whenever possible in all procedures.
